Thermal inactivation of Phytophthora capsici oospores.

چکیده
BACKGROUND Phytophthora capsici is a major fungal plant pathogen that causes root and crown rot of pepper crops and its oospores are the most resistant propagules. OBJECTIVES To evaluate the effect of different temperature regimes and exposure times on the survival of P. capsici oospores. METHODS Thermal inactivation treatments simulated field conditions, through the use of different constant and cycling temperature regimes, in moistened sterilized soil (15-53 °C) and sterilized water (45-53 °C). The plasmolysis method evaluated oospore viability. Relationships between oospores viability and exposure time were statistically determined by linear regression. Interpolation was used to calculate the estimated times required to kill a determined percentage of the population. RESULTS The required time to reduce P. capsici oospores viability decreased with increasing temperatures. Times required to kill 100% of oospores were 199-22-6.6-4.7-1.0 hours at 40-45-47.5-50-53°C respectively in moistened soil and 31-1.0-0.2 hours at 45-50-53 °C in water. Oospores were scarcely affected at temperatures ≤ 35 °C. With 1,680 hours at 15-35 °C, oospores survival in soil ranged from 88 to 36%. The 4 hours-40 °C regime killed 100% of oospores after 28days, while the 5 hours-35°C regime after 70 days killed only 75%. Time required to achieve total oospores death was remarkably shortened in water when compared with moistened soil. CONCLUSIONS The developed models can be used to predict survival values at any exposure time with constant temperatures ranging from 40 to 53 °C in moistened soil and from 45 to 53 °C in water. The weakening of P. capsici oospores under sublethal heating, is a useful observation that can be applied for pathogen control with solarization.

Determination of viability of Phytophthora capsici oospores with the tetrazolium bromide staining test versus a plasmolysis method.
BACKGROUND Oospores are the most persistent propagules of Phytophthora capsici. The viability of oospores is determined by staining methods such as the tetrazolium bromide (MTT) test. OBJECTIVES The aim was to assess the MTT test and a plasmolysis method for their utility in determining viability of oospores of P. capsici. METHODS For either procedure the reactions of non-treated (viable) a...
متن کاملMefenoxam Insensitivity and the Sexual Stage of Phytophthora capsici in Michigan Cucurbit Fields.
ABSTRACT The potential for outcrossing, occurrence of oospores, and inheritance of mefenoxam sensitivity was assessed in naturally occurring populations of Phytophthora capsici. Between 1997 and 1998, 14 farms were sampled, with 473 isolates recovered from cucurbit hosts and 30 from bell pepper. The A1 and A2 compatibility types were recovered in a roughly 1:1 ratio in 8 of 14 farms with sample...
متن کاملTemporal Genetic Dynamics of an Experimental, Biparental Field Population of Phytophthora capsici
Defining the contributions of dispersal, reproductive mode, and mating system to the population structure of a pathogenic organism is essential to estimating its evolutionary potential. After introduction of the devastating plant pathogen, Phytophthora capsici, into a grower's field, a lack of aerial spore dispersal restricts migration. Once established, coexistence of both mating types results...
متن کاملThe Spatiotemporal Genetic Structure of Phytophthora capsici in Michigan and Implications for Disease Management.
Root, crown, and fruit rot caused by Phytophthora capsici Leonian is a limiting factor for the production of peppers, tomatoes, and cucurbit crops in Michigan and the United States. Like many species in the genus Phytophthora, P. capsici has the potential for rapid polycyclic disease development from a limited amount of initial inoculum (6). P. capsici produces caducous sporangia that can be sp...
متن کاملBiologically active Phytophthora mating hormone prepared by catalytic asymmetric total synthesis.
A Phytophthora mating hormone with an array of 1,5-stereogenic centers has been synthesized by using our recently developed methodology of catalytic enantioselective conjugate addition of Grignard reagents. We applied this methodology in a diastereo- and enantioselective iterative route and obtained two of the 16 possible stereoisomers of Phytophthora hormone alpha1. These synthetic stereoisome...
